No. Paul is quoting Malachi 1, written roughly 1,400 years after the brothers lived. By Malachi's day, "Esau" meant the nation Edom — not the individual.

In ancient Near Eastern covenant language, love/hate is a chosen / not-chosen idiom for covenant purpose, not personal animosity. Jesus uses the same idiom in Luke 14:26 when He says we must "hate" father and mother to follow Him.
